The Martyrs play Taunton Town away at their Viridor Stadium with £3,250 prize money on offer for the winners.
Manager Gavin Williams and his players lifted confidence within the group thanks to a 3-1 home win against Stratford Town in midweek thanks to goals by Ian Traylor, Kerry Morgan and Eliot Richards.
Taunton, nicknamed the Peacocks, are top of the Southern League West table after an impressive start to their season.
They progressed to the first qualifying round of the FA Trophy with a 2-1 victory against AFC Totton in a preliminary round replay earlier this month.
Captain Matt Wright has been influential since his arrival with 28 goals in 49 appearances and the prolific striker is enjoying another competitive season.
Merthyr are still in touch with the leading group at Premier Division level and are keen to stamp their authority on Taunton in the Buildbase FA Trophy.
Kyle Patten and Ian Traylor sat out training at the Loadlok Community Stadium this week, but both are expected to be fit for the trip to Somerset on Saturday.
Big striker Ryan Prosser took part in light training and is encouraged by his progress, but his back injury is still being monitored.
If a replay against Taunton is needed it will be played at Merthyr Town next Tuesday, October 31.
The next League fixture for the Martyrs is against Redditch United away on Saturday, November 4, while they then have back-to-back home matches against Banbury United and Basingstoke.
Banbury travel to Merthyr on Saturday, November 11, while Basingstoke Town are visitors on the following Tuesday.
